Whole Genome Sequencing and Safety Evaluation of Lactiplantibacillus plantarum SC-1

  1. Lactiplantibacillus plantarum SC-1, a strain able to promote the production of ergosterol by Monascus isolated from naturally fermented Chinese sauerkraut, was evaluated for genetic characteristics and application safety. Whole genome sequencing and Kyoto Encyclopedia of Genes and Genomes (KEGG) and gene ontology (GO) annotation showed that the functional genes of strain SC-1 are involved in pathways including amino acid synthesis, carbon metabolism, genetic information regulation, and environmental stress response. The carbohydrate-active enzymes (CAZy) of this strain were predominantly glycoside hydrolases and glycosyltransferases, indicating that the strain can metabolize carbohydrates. Acid and bile salt tolerance assays demonstrated that the survival rate of SC-1 was (68.89 ± 3.91)% in an acidic environment (pH 2.5) and (50.30 ± 4.25)% at 0.3% (m/m) bile salt content, respectively. In addition, SC-1 exhibited γ-hemolysis (non-hemolytic activity) and it was sensitive to common antibiotics such as penicillin but showed resistance to several aminoglycoside antibiotics. SC-1 had no significant virulence and its resistance genes posed no transfer risk.

摘要: 对一株分离自天然酸菜?可促进红曲霉产麦角甾醇的植物乳植杆菌(Lactiplantibacillus plantarum)SC-1 的遗传特征与应用安全性进行研究?通过对SC-1菌株进行全基因组测序,结合京都基因与基因组百科全书及基因本体论数据库注释,得出SC-1菌株功能基因参与氨基酸合成?碳代谢?遗传信息调控及环境应激响应等通路?碳水化合物活性酶数据库注释表明,SC-1菌株的碳水化合物活性酶以糖苷水解酶和糖苷转移酶为主,具备碳水化合物代谢能力?耐酸?耐胆盐实验表明,SC-1菌株在pH 2.5酸性环境和0.3%(m/m)胆盐环境中的存活率分别达(68.89±3.91)%和(50.30±4.25)%?此外,SC-1菌株为γ-溶血,无溶血性,对青霉素等常用抗生素敏感,对部分氨基糖苷类抗生素耐药?毒力因子与抗生素抗性基因分析表明,SC-1菌株无明显毒力作用,抗性基因无转移风险?
