β-乳球蛋白与二十二碳六烯酸互作效应对β-乳球蛋白致敏性机制的影响

The impact of the β-lactoglobulin and Docosahexaenoic Acid interaction on the sensitization mechanism of β-lactoglobulin

摘要: β-乳球蛋白(β-lactoglobulin,β-LG)被普遍认为是牛乳中的主要过敏原,约有82.0%牛乳过敏人群对β-LG产生强烈的过敏反应。本实验以β-LG为原料,采用分子模拟对接法、同步荧光光谱法、三维荧光光谱法、圆二色谱法和间接竞争酶联免疫吸附测定法等探究二十二碳六烯酸(Docosahexaenoic Acid,DHA)与β-LG互作效应及其对β-LG致敏性的影响。结果表明,DHA主要通过疏水作用力与β-LG结合,且形成稳定的复合物。且DHA与β-LG结合后,改变β-LG的二级结构,其中β-LG的β-折叠含量增加约30%。同时结合后β-LG极性增加,疏水性减弱,致敏性降低约为30%。本研究通过解析DHA与β-LG互作效应对其致敏性的影响,为开发低致敏性乳制品提供理论基础。

Abstract: β-lactoglobulin (β-LG) is widely recognized as a major allergen in cow's milk, and about 82.0% of cow's milk-allergic people have strong allergic reactions to β-LG. In this experiment, molecular simulation docking method, synchronous fluorescence spectroscopy, three-dimensional fluorescence spectroscopy, circular dichroism and Elisa method were used to investigate the interaction effect of Docosahexaenoic Acid (DHA) with β-LG and its influence on the allergenicity of β-LG. The results showed that DHA binds to β-LG mainly through hydrophobic forces and forms stable complexes. And the binding of DHA to β-LG changed the secondary structure of β-LG, in which the β-turn content of β-LG increased by about 30%. Meanwhile, the polarity of β-LG increased and hydrophobicity was weakened after binding, and the sensitizing property was reduced by about 30%. This study provides a theoretical basis for the development of hypoallergenic dairy products by analyzing the influence of the interaction effect between DHA and β-LG on their allergenicity.
