不同泌乳期山羊乳与牛乳类固醇激素含量的差异性分析

Analysis of differences in steroid hormone content between goat milk and cow milk at different lactation stages

摘要: 摘要:【目的】为了了解不同泌乳期山羊乳与牛乳中类固醇激素含量的代谢差异。【方法】选取健康状况良好、饲养条件一致的处于泌乳初期(泌乳2 d),泌乳盛期(泌乳90 d )和泌乳末期(泌乳220 d)的关中奶山羊和荷斯坦奶牛各6只,通过液相色谱质谱联用LC-MS/MS)检测不同泌乳期乳汁中类固醇激素水平含量。检测的色谱条件流动相为A(氟化铵水溶液)和B(甲醇)、正离子喷雾电压为5500V;负离子喷雾电压为-4500V,离子源温度为550℃,柱温为45℃条件下;定量利用三重四极杆质谱的多反应检测(MRM)模式进行分析。结合电喷雾离子源负离子模式检测。【结果】结果表明,以24种靶标为参照,从山羊乳与牛乳各泌乳期共检测出9种类固醇类激素,分别是17a-羟基孕烯醇酮、17a-孕烯醇酮、表睾酮、孕酮、雄稀二酮、睾酮、雌激素、皮质醇、可的松。且在泌乳初期山羊乳与牛乳之间含量差异较大的激素是17a-羟基孕烯醇酮、雌激素、皮质醇(P<0.05);泌乳盛期含量差异较大的激素是17a-羟基孕烯醇酮、孕酮、皮质醇、可的松(P<0.05);泌乳末期山羊乳与牛乳含量差异较大的激素是17a-羟基孕烯醇酮、孕酮、睾酮可的松(P<0.05)。【结论】山羊乳与牛乳中类固醇激素含量的差异可能是由于妊娠状态的不同导致的,研究结果为功能性乳制品的设计和类固醇激素代谢疾病的预防提供理论依据。

Abstract: Abstract: 【Purpose】To understand the metabolic differences in steroid hormone content between goat milk and cow milk at different lactation stages.【Method】Select 6 Guanzhong dairy goats and 6 Holstein cows each in the early lactation stage (2 days of lactation), peak lactation stage (90 days of lactation), and late lactation stage (220 days of lactation) with good health and consistent feeding conditions. Detection of steroid hormone levels in milk at different lactation stages by liquid chromatography-mass spectrometry (LC-MS/MS). The chromatographic conditions for detection were as follows: mobile phases A (ammonium fluoride aqueous solution) and B (methanol), positive ion spray voltage 5500V; The negative ion spray voltage is -4500 V, the ion source temperature was 550 ℃, and the column temperature was 45 ℃; Quantitative analysis using the multiple reaction detection (MRM) mode of triple quadrupole mass spectrometry. Combined with negative ion mode detection of electric spray ion source. 【Results】The results showed that a total of 9 types of steroid hormones were detected from goat milk and cow milk at different lactation stages, using 24 different targets as reference. These hormones were 17α-hydroxypregnenolone, 17α-pregnenolone, 17-Epi-Testosterone, progesterone, androstenedione, testosterone, estrogen, cortisol, and cortisol, respectively. The hormones with significant differences in content between goat milk and cow milk during the early lactation stage are 17a hydroxypregnenolone, estrogen, and cortisol (P<0.05); The hormones with significant differences in content during the peak lactation period were 17a hydroxyprogesterone, progesterone, cortisol, and cortisol (P<0.05); The hormones with significant differences in content between goat milk and cow milk in the late lactation stage are 17a hydroxypregnenolone, progesterone, and testosterone cortisone (P<0.05). 【Conclusion】The difference in steroid hormone levels between goat milk and cow milk may be due to differences in pregnancy status, the research results provide theoretical basis for the design of functional dairy products and the prevention of steroid hormone metabolism diseases.
