Determination of Casein Phosphopeptides in Powdered Formulae for Infants and Young Children by High Performance Liquid Chromatography

高效液相色谱法检测婴幼儿配方乳粉中酪蛋白磷酸肽

Abstract: An analytical method was developed for the determination of casein phosphopeptides (CPP) in powdered formulae for infants and young children by high performance liquid chromatography. Three pretreatment methods and two operating conditions were optimized. The results showed that the optimal pretreatment conditions were as follows: transferring samples to a 50 mL volumetric flask after pH adjustment to 4.6 with 1 mol/L hydrochloric acid, and then filtering them through a filter paper. The chromatographic separation was carried out by gradient elution using 0.05% trifluoroacetic acid as mobile phase A and 0.05% acetonitrile as mobile phase B. The spiked recovery was between 94.50% and 95.45%, good linearity was observed with R2 = 0.999 8, and the detection limit was 2.0 mg/100 g. The method had the advantages of simple sample pretreatment as well as good stability and reliability, and the results from this method met the requirements in terms of linearity, recovery, accuracy, precision and detection limit. Thus it could be used to determine the content of CPP in powdered formulae for infants and young children.

摘要: 建立一种高效液相色谱法检测婴幼儿配方乳粉中酪蛋白磷酸肽(casein phosphopeptides,CPP)含量 的方法。对3 种样品前处理方法及2 种上机条件进行优化,确定最优条件。结果表明:最优前处理条件为用盐 酸(1 mol/L)调节乳粉样品溶液pH 4.6后用水转移至50 mL容量瓶中,用滤纸过滤后作为待测液;上机检测时 用0.05%三氟乙酸-0.05%乙腈作为流动相体系进行梯度洗脱;样品加标回收率为94.50%~95.45%,线性良好 (R2=0.999 8),方法检出限为2.0 mg/100 g。该方法样品前处理简单、稳定性和可靠性好,检测结果满足线性、精 密度、准确度及检出限要求,可用于婴幼儿配方乳粉中CPP含量的测定。
