高效液相色谱法检测乳制品中硫氰酸根含量

Abstract: A method for detecting thiocyanate in dairy products by high-performance liquid chromatography (HPLC) is described. The proteins in the samples were precipitated by the addition of acetonitrile, then the filtrate was separated on a strong anion exchange (SAX) column using a mobile phase made up of a mixture of sodium dihydrogen phosphate solution and methanol. The target compound was detected with a UV detector at 218 nm. The results showed that the calibration curve displayed a good linear relationship in the range of 0.2–100 mg/kg. The recovery rate of thiocyanate was 98.5%– 113.0%.The method is simple with good accuracy and stability for detecting thiocyanate in dairy products.

摘要: 建立乳制品中硫氰酸根的高效液相色谱分析方法.样品经乙腈沉淀蛋白,以磷酸二氢钠磷酸缓冲液-乙腈溶液为流动相,强阴离子交换(SAX)色谱柱分离,紫外检测器218 nm检测.结果表明:在硫氰酸根0.2~100 mg/kg范围内呈良好的线性关系,回收率为98.5%~113.0%.本方法用于乳制品中硫氰酸根的测定,操作简单、定量准确、稳定性好.
