Determination of β-Sitosterol in Milk by GC-MS

气相色谱-质谱联用法测定牛乳中β-谷甾醇

Abstract: A gas chromatography-mass spectrometry (GC-MS) method was developed for determining β-sitosterol in milk by optimizing important sample pretreatment parameters such as saponification temperature, extraction solvent, extraction number, shaking method during extraction and rotary evaporation temperature. The feasibility of the developed method was examined. This method exhibited a wide linear range and could meet the requirements for routine quantitative analysis of β-sitosterol in milk samples with high precision, good repeatability, accurate results and high reliability.

摘要: 对气相色谱-质谱联用法测定牛乳中β-谷甾醇的样品前处理步骤及方法可行性进行研究及优化设计.通过研究多个关键单因素对皂化温度、萃取剂选择、萃取次数及萃取过程中的振摇方式、旋转蒸发温度等因素的影响,最终得到了适用于对生鲜乳的前处理方法及步骤;并且研究气相色谱-质谱联用法测定生鲜乳中β-谷甾醇含量的可行性.结果表明:采用的气相色谱-质谱联用检测方法线性范围较宽,完全能满足常规牛乳试样中β-谷甾醇各质量浓度范围内的定量分析.该方法分析精密度高、重复性较好、分析结果准确、可靠性高.
